What Does This Test Measure?

The panel measures testosterone, estradiol, progesterone, DHEAS and cortisol, with cortisol tested twice to compare morning and bedtime levels.

Hormone What it adds to your assessment
Testosterone Information relevant to concerns about libido, energy, strength and motivation
Estradiol (E2) An assessment of oestrogen levels alongside testosterone
Progesterone Additional information about your sex hormone profile
DHEAS Measurement of an adrenal androgen that contributes to the production of other sex hormones
Morning cortisol Your cortisol level at the beginning of the waking day
Bedtime cortisol Your cortisol level before sleep, allowing comparison with the morning result

How Can This Test Help You?

Explore hormone levels alongside your symptoms

The results provide a starting point for investigating concerns such as low libido, fatigue and changes in mood or physical performance. Assessing several hormones together offers more context than a testosterone result alone. ZRT includes sex hormone and cortisol testing among its options for men experiencing these concerns.

Understand morning and bedtime cortisol

If you struggle to get going in the morning or feel unusually alert at bedtime, two cortisol measurements provide information about these different parts of your day. Cortisol normally follows a daily rhythm; this panel compares its morning and bedtime levels, while a four-point profile provides additional daytime detail.

Turn results into useful next steps

Specialist interpretation helps you understand your results alongside your symptoms, medications and current treatments. This can support more focused discussions about sleep routines, stress management, exercise recovery and whether further assessment would be helpful.

Create a baseline for future comparison

Your report provides a record of your hormone levels. Where repeat testing is appropriate, it can help you and your practitioner assess changes over time.

Who Should Consider This Test?

This profile may be of interest to men experiencing:

  • Reduced libido or changes in sexual function
  • Persistent fatigue or reduced stamina
  • Poor sleep or difficulty winding down at night
  • Reduced strength or disappointing exercise recovery
  • Changes in body composition
  • Low motivation, irritability or difficulty concentrating
  • Symptoms they would like to explore alongside their hormone levels

It is also an option for people seeking a hormone baseline or discussing hormone treatment monitoring with their practitioner. These concerns can have several causes, so results are most useful when considered alongside your wider health history.

Why Choose Saliva Hormone Testing?

Saliva collection is simple, non-invasive and convenient for sampling at different times of day. ZRT offers LC-MS/MS saliva profiles and describes their sensitivity as particularly useful when measuring low oestrogen concentrations, including those found in men and children.

Saliva testing provides information about hormones present in saliva, including unbound steroid hormones. Its usefulness depends on the hormone being assessed and the clinical question; it complements other testing options.

Frequently Asked Questions

Does this test measure five or six hormones?
It measures five different hormones. Cortisol is measured twice, giving six individual results: testosterone, estradiol, progesterone, DHEAS, morning cortisol and bedtime cortisol.

How many saliva samples do I need?
Two samples on the same day: one in the large tube upon waking and one in a small tube before bed. Your kit may contain additional tubes that are not required for S-145.

How does this differ from Male Saliva Hormone Profile I?
Profile I, S-124, measures the same five hormones using one morning sample. S-145 adds a bedtime cortisol measurement, allowing comparison between the start and end of your waking day.

Can I use this test if I am on TRT?
It may be suitable, depending on how your testosterone treatment is administered. Follow Section 3 of the collection instructions for treatment timing and contamination prevention. Continue prescribed hormones as directed; the instructions recommend a different sample type for troches or sublingual hormone therapy.

Is this test suitable for adults and children?
Yes. The collection method is suitable for adults and children who can provide the required saliva volume. For a child, include their age and relevant symptoms so the results can be considered in the appropriate context.

Will the results explain my fatigue or low libido?
They can help show whether the measured hormone levels warrant further attention. Specialist interpretation connects the laboratory findings with the information you provide and helps guide your next steps.

Does the price include laboratory analysis?
Yes. Laboratory analysis and specialist interpretation are included. You pay separately for returning your samples.

How long is the kit valid for?
Use the kit within 12 months of purchase.

Where can I find the full collection instructions?
Download the Saliva Collection Instructions PDF. Follow the S-145 two-sample schedule.
