Saliva kits can require different numbers of tubes and collection times, depending on the panel. Start by matching the test code on your requisition to the Saliva Collection Instructions PDF. Choose the right collection day
The instructions say men, children and non-menstruating women can collect on any day of the month. For menstruating women, timing depends on the panel. If testing FSH for fertility reasons, the PDF says to collect on cycle day 3. If testing estradiol (E2), progesterone (Pg) or testosterone (T), it gives these examples for regular cycles: 23–26 days, collect on days 17–19; 27–30 days, days 21–23; 31–34 days, days 25–27. For longer or shorter cycles, it says to collect 5–7 days before the expected next period. For infrequent or unpredictable cycles, collect on a non-bleeding day. Day 1 is the first day of period flow, not spotting. For other panels, the instructions say any day of the month.
Prepare before collection
Check your test code and collection timetable first, because the number and timing of saliva samples vary by panel. The PDF advises avoiding anti-aging or anti-wrinkle facial creams and biotin (vitamin B7) hair and nail formulas for three days beforehand. If you take a prescribed or medically advised supplement, check with your healthcare professional before changing it.
On collection day, do not eat or drink anything except water, or brush your teeth, before the first sample and for two hours before collections later in the day. Rinse your mouth with water and wait five minutes before beginning the first collection. The night before, stay hydrated but avoid drinking excessively.
Collect and label each sample
- Wash and dry your hands. Begin the first sample within 30 minutes of waking, using the large 10 mL tube if your panel requires it.
- Let saliva pool in your mouth, then transfer it to the tube. Fill the tube at least halfway, not counting bubbles. The instructions say collection usually takes 15–30 minutes.
- Screw the cap on securely. Write your name, collection date and time on the tube label, then return it to its bag.
- Repeat at the other times on your panel timetable, using the smaller 5 mL tubes if provided and required.
Hormone-use notes
List all current medicines and hormones on the requisition. The PDF says not to stop hormone use and to continue as prescribed, while also giving specific timing and contamination precautions for different hormone types. For example, it advises avoiding bare-hand application of topical hormones for at least two days before collection and using gloves or an applicator. It says troches and sublingual therapies are not recommended for saliva hormone testing; discuss alternatives with your healthcare professional. Ask your provider about timing if you use steroids or if the instructions conflict with your treatment plan.
Store and return the samples
If a saliva sample will not be shipped within 24 hours, the instructions say to freeze it. They state that samples remain stable under average shipping conditions, including weekends and holidays. Complete all paperwork fully, pack the samples and requisition in the kit box, then use the supplied return packaging and appropriate carrier.
